HPLC separates; the detector records a signal

High-performance liquid chromatography moves a sample through a column. Components interact differently with the column and can emerge at different times. The chromatogram plots detector response against time. A peak is a measured signal, not a photograph of a pure molecule.

For a peak-area purity calculation, the laboratory compares the target area with the included peak areas. The detector, separation conditions and integration rules influence that calculation.

Why one clean peak can be misleading

An impurity that gives little response under the chosen detector conditions can also be understated. This is why “HPLC tested” needs a method and interpretation. Method selectivity addresses whether the target can be measured despite other components. FDA / ICH Q2(R2): analytical validation.

LC-MS adds mass information

LC-MS combines chromatography with mass spectrometry, which measures mass-to-charge ratio (m/z). A peptide can carry several charges and appear at more than one m/z value. The laboratory interprets those signals to assess the expected molecular mass and other identification criteria.

Some compounds share a mass or have closely related structures. A mass match is not proof of every detail of sequence, stereochemistry or composition.

Quantity needs calibration

A larger peak can indicate more material under comparable conditions, but the graph alone does not establish milligrams per vial. Quantification needs appropriate calibration and a documented preparation basis. Different compounds may produce different detector responses at the same amount.
